Protective role of One Period of Submaximal Swimming Exercises and Nigella Sativa Nano-capsules Injection on Cyclin D1 Gene Expression in Lung Tissue of Wistar Rats following the Exposure to Carcinogen NNK

Abstract
It is believed that exercise and dietary supplements are capable of affecting lung tissue exposed to carcinogens in cigarette smoke to some extent. The present study aims at exploring the effect of 12 weeks of swimming exercises and injection of Nigella Sativa nano-capsules on cyclin D1 gene expression in the lung of rats following exposure to carcinogen NNK.
85 Wistar rats weighing averagely105 ± 26 g were put into 9 control groups (n = 6), distilled water injection (n = 10), NNK injection (n = 10), swimming exercise (n = 10), supplement (n = 9), NNK + swimming practice (n = 10), NNK + supplement (n = 10), supplement + swimming practice (n = 10) and supplement + NNK + swimming (n = 10) groups. Swimming exercise was done five days a week for twelve weeks. NNK and Nigella Sativa nano-capsules were injected subcutaneously once a week for 12 weeks at a dose of 12.5 mg and 125 µg / kg of rats, respectively; distilled water was injected to the vehicle group once a week by the same dose and under the same condition. 48 hours after the last exercise session and injection, lung tissue samples were gathered and cyclin D1 gene expression was performed using Real-Time PCR technique. One way ANOVA and Tukey post hoc tests were used for data analysis at the level of  ≤ 0/05.
According to the results, 12 weeks of swimming exercises in NNK + swimming group reduced the expression of cyclin D1 gene (P = 0.0002) compared to NNK group. In addition, 12 weeks of Nigella Sativa nano-capsules injection in the supplement + NNK compared to the NNK group decreased the expression of cyclin D1 gene (P = 0.003). In addition, the interaction of swimming and supplemental exercise in swimming + supplement + NNK compared to NNK group reduced the expression of cyclin D1 gene (P = 0.001).
It seems that swimming exercises and Nigella Sativa nano-capsules could be effective in preventing pathways leading to NNK-induced tumor in the cell cycle.
